Asimismo, esta lectina tuvo efecto hemaglutinante en suspensiones de glóbulos rojos de los grupos sanguíneos ABO. Las suspensiones de las levaduras tuvieron una concentración de 2,4 ×108 comparada con el tubo 8 de Mac Farland. El efecto aglutinante se demostró tanto en extracto crudo como en pre- purificado con alcohol de 95º. Cuando la suspensiones de Candida albicans, Saccharomyces cerevisiae o Levadura roja son tratadas con glicina – HCl y sometidas a<br />ebullición por 15 minutos pierden la capacidad de aglutinarse con el extracto crudo de semillas de “ñuña” y con<br />extracto pre – purificado con alcohol. Las especies de levaduras fueron cultivadas en agar sabouraud y cosechada<br />con PBS. Los extractos de “ñuña” se obtuvieron a partir de semillas que fueron molidas a polvo fino, tamizados y extraída la lectina con ácido clorhídrico 0.1N, centrifugada y el sobrenadante (extracto crudo) fue precipitado con alcohol, el precipitado fue separado por centrifugación, y redisuelto con PBS (extracto pre purificado). Se determinó la concentración de proteínas por los métodos de Biuret y Lowry.<br />Palabras clave: Lectinas, Extracción, Purificación, Phaseolus vulgaris L. var. ñuña “ñuña”, Candida albicans,<br />Saccharomyces cerevisiae y levadura roja.</p><p>ABSTRACT<br />Phaseolus vulgaris L. var. ñuña “ñuña” the lectins which was obtained by acid extraction, showed a binding agglutinating effect against Candida albicans suspensionin PBS to at room temperature and observed after one hour. C. albicans suspension had a concentration of 2, 4 *108 compared with Farland Mac tube 8. Binding effect demonstrated so much in crude extract as pre-purified with alcohol of 95º. “Ñuña” seeds were worn out to fine dust, sifted and extracted with hydrochloric acid 0.1N, centrifuged and the supernatant (crude extract) it was precipitated with alcohol, the precipitated one was separated by centrifugation and suspended with PBS (extract pre purified). The protein concentration was determined by Candida albicans was cultivated in agar sabouraud and harvested with PBS. When Candida albicans is treated with glicina - HCl and submitted boiling by 15 minutes, agglutination did not present the crude extract of seeds of “ñuña” or pre-purified extract with alcohol.<br />Key words: Lectins, Extraction, Purification, Phaseolus vulgaris L. var. ñuña “ñuña”, Candida albicans, Saccharomyces cerevisiae.</p> ER -
